چکیده

The axial tissues of the vertebrate embryo is a term that refers collectively to the floor plate (a cell population that lies at the ventral floor of the neural tube) and the notochord (which is a mesodermal rod of cells located immediately ventral to the floor plate). The notochord and floor plate both secrete the morphogen signal Sonic Hedgehog (Shh) that patterns the dorsoventral axis of the neural tube. Moreover the ability of both tissues to induce floor plate differentiation is mediated by Shh. These two tissues are believed to derive from a common progenitor pool which is located in Hensen’s node in the chick embryo. Hensen’s node develops at the anterior tip of the primitive streak. Some of these progenitor cells remain in the node as resident stem cells. It is unclear how the progenitor cells in Hensen’s node become committed to either the notochord or floor plate cell fate. The Notch signalling pathway is a key player in binary cell fate choice in numerous other developmental contexts in the developing vertebrate and invertebrate embryo and we have found this pathway plays a role in the cell fate decision that occurs in the axial tissue precursors in Hensen’s node. We also show that Notch signalling modulates the response of neural cells to the morphogen Shh.

Analysis of FGF-Dependent and FGF-Independent Pathways in Otic Placode Induction

The inner ear develops from a patch of thickened cranial ectoderm adjacent to the hindbrain called the otic placode. Studies in a number of vertebrate species suggest that the initial steps in induction of the otic placode are regulated by members of the Fibroblast Growth Factor (FGF) family, and that inhibition of FGF signaling can prevent otic placode formation. To better understand the genet...

Integrin-α5 Coordinates Assembly of Posterior Cranial Placodes in Zebrafish and Enhances Fgf-Dependent Regulation of Otic/Epibranchial Cells

Vertebrate sensory organs develop in part from cranial placodes, a series of ectodermal thickenings that coalesce from a common domain of preplacodal ectoderm. Mechanisms coordinating morphogenesis and differentiation of discrete placodes are still poorly understood. We have investigated whether placodal assembly in zebrafish requires Integrin- α5 (itga5), an extracellular matrix receptor initi...

A conserved role for FGF signaling in chordate otic/atrial placode formation.

The widely held view that neurogenic placodes are vertebrate novelties has been challenged by morphological and molecular data from tunicates suggesting that placodes predate the vertebrate divergence. Here, we examine requirements for the development of the tunicate atrial siphon primordium, thought to share homology with the vertebrate otic placode. In vertebrates, FGF signaling is required f...

Fgf3 and Fgf10 are required for mouse otic placode induction.

The inner ear, which contains the sensory organs specialised for audition and balance, develops from an ectodermal placode adjacent to the developing hindbrain. Tissue grafting and recombination experiments suggest that placodal development is directed by signals arising from the underlying mesoderm and adjacent neurectoderm. In mice, Fgf3 is expressed in the neurectoderm prior to and concomita...

FGF Signaling Regulates Cytoskeletal Remodeling during Epithelial Morphogenesis

Changes in the cytoskeletal architecture underpin the dynamic changes in tissue shape that occur during development. It is clear that such changes must be coordinated so that individual cell behaviors are synchronized; however, the mechanisms by which morphogenesis is instructed and coordinated are unknown. After its induction in non-neural ectoderm, the inner ear undergoes morphogenesis, being...
